High sequence coverage is crucial for epitope mapping as the epitope may only contain a few amino acid residues. The traditional bottom-up HDX-MS workflow often results in very small peptides too hydrophilic to be retained on LC columns and is unable to detect post-translationally modified peptides such as glycopeptides. The FineMapping platform was developed to effectively tackle these limitations. PD-1 is an important drug target for biologics development, but it is heavily glycosylated. Epitope mapping on PD-1 by traditional HDX-MS and other methods only provides incomplete information.
